Francois Louw will start at number six for the Springboks when they take on Wales in Durban on Saturday (kick-off 16.00, live on Sky Sports 1).
There are six changes to the matchday squad from the team which comprehensively beat the World XV last weekend in Cape Town, four of which are positional shifts.
Louw is one of two Aviva Premiership players featured in the squad, with Saracens hooker Schalk Brits on the bench. Wales's tour of South Africa began on Tuesday with a convicing 12-34 defeat of Eastern Province Kings, with Bath Rugby loosehead Paul James playing an important role in the dominance of the Welsh pack.
Springboks coach Heyneke Meyer believes Wales will pose a stern challenge to his side on Saturday.
“They started their tour well [and] are always fired up and ready,” said Meyer, who formerly oversaw Leicester Tigers. “Wales have an experienced coaching team and they were the Six Nations champions in 2012 and 2013, as well as the back-bone of the British & Irish Lions team which beat Australia last year, so this will be a massive challenge in our first Test of the season.”
